Genetic characteristics of rabies virus cause endemic in Quang Nam and Quang Ngai, 2016-2017

Rabies has been endemic in Viet Nam for years due to its association with dogs and it has been the serious public health concern in Vietnam. Quang Nam, Quang Ngai provinces located in Centre of Vietnam have constantly reported for rabies deaths for years. The genetic characterization of rabies viruses (RABV) in this endemic area is limited. To develop an effective control strategy, it is important to understand the transmission patterns of the RABV. The objectives of this study were to describe genetic characteristics of the viruses in Quang Nam, Quang Ngai provinces in 2016- 2017. Three viruses obtained from two patients (H103; H105) and cow (C201) using RT-PCR and DFA. Phylogenetic analysis of their partial nucleotide sequences of Nucleoprotein (N) gene was performed using the maximum-likelihood method in conjunction with other RABVs isolated in Vietnam and neighboring countries collected in previous studies. The results showed that three viruses belonged to RABVs genotype 1, sub-group 1B consisted of viruses circulated in Highland and South of Vietnam. The phylogenetic analysis revealed close relationship between viruses collected in patient and cow in Quang nam 2016, 2017. They formed a cluster together with RABVs from dogs collected in 2009 in South of Vietnam. The virus isolated in patient in Quang Ngai 2016 formed a distinct cluster from Quang Nam viruses. It was closely associated with viruses identified previously from dogs in Gia Lai province in 2009. A single dominant aa of A84 or A135 uniquely detected in the South and Highland of Vietnam was found in identified viruses Our results suggest that these viruses may have originated from a common ancestor and spread between difference South of Centre, South and Highland of Vietnam. This is the first data provided understanding of the relatedness and genetic characteristics of RABVs in Center of Vietnam
